Cagrilintide (AM833) is a novel long-acting acylated amylin analog developed by Novo Nordisk. It acts as a non‑selective agonist of amylin receptors (AMY1R, AMY2R, AMY3R) and the calcitonin receptor (CTR). The peptide features a C20 fatty diacid moiety attached via a γ‑glutamic acid linker, which enables albumin binding and extends the half‑life to approximately 7‑8 days, supporting once‑weekly administration.

Unlike GLP‑1‑based therapies, cagrilintide targets amylin pathways in the brain to induce satiety and reduce food intake, offering a distinct mechanism of action that can be used alone or in combination with semaglutide (CagriSema).
